五界系統(tǒng)是以細(xì)胞結(jié)構(gòu)分化的等級以及和光合、吸收、攝食這三種主要營養(yǎng)方式有關(guān)的組織類型為基礎(chǔ)的。六界:加上病毒界。 Woese用寡核苷酸序列編目分析法對60多株細(xì)菌的16SrRNA序列進(jìn)行比較后,驚奇地發(fā)現(xiàn):產(chǎn)甲烷細(xì)菌完全沒有作為細(xì)菌特征的那些序列,于是提出了生命的第三種形式--古細(xì)菌(archaebacteria)。隨后他又對包括某些真核生物在內(nèi)的大量菌株進(jìn)行了16SrRNA(18SrRNA)序列的分析比較,又發(fā)現(xiàn)極端嗜鹽菌和極端嗜酸嗜熱菌也和產(chǎn)甲烷細(xì)菌一樣,具有既不同其他細(xì)菌也不同于其核生物的序列特征,而它們之間則具有許多共同的序列特征。于是提出將生物分成為三界(Kingdom)(后來改稱三個域):古細(xì)菌、真細(xì)菌(Eubacteria)和真核生物(Eukaryotes)。1990年,他為了避免把古細(xì)菌也看作是細(xì)菌的一類,他又把三界(域)改稱為:Bacteria(細(xì)菌)、Archaea(古生菌)和Eukarya(真核生物),并構(gòu)建了三界(域)生物的系統(tǒng)樹。
